PES 2016 akan hadir lebih awal dibanding pendahulunya PES 2015. Jika rilis Pro Evolution Soccer 2015 pada 13 November 2015, maka PES 2016 akan dirilis bulan September atau paling lambat pertengahan Oktober tahun ini. Hal ini seperti dikatakan oleh Direktur Konami wilayah Eropa Selatan, Stéphanie Hattenberger :
"I know the release date of PES 2016, but I cannot reveal it today. The only thing I can say is that the game will be released earlier than PES 2015."
Pada Pro Evolution Soccer 2016 akan menampilkan Neymar Jr. sebagai pengganti Mario Gotze. Sang bintang Brazil itupun kemudian berkomentar : “This is a real honor for me to be cover star for PES 2016. The series has long been praised for its sheer level of control and utter realism and I am delighted to be working with such a talented team as Konami. I am fascinated by the huge amount of work involved in putting together the game and ensuring the world’s top players look and play just like the real things, and cannot wait to see myself in action as soon as possible.”
